Over the years I have developed a bad seal which drains my main display tank from an overflow box into PVC drain pipes.  I get a slow leak.    I have used silicone multiple times and it will usually slows down and eventually stops.  If the drain pipe gets bumped it can break the seal and I have to start over.  Is there a better/faster way then schlopping silicone over a wet seal?  (This is one of my two drain pipes. The other one on the other side of the tank does not leak.)

What I would do is drain the overflow box and redo the bulkhead. Really easy to do. And a much better fix then trying to silicon over a problem.

This, 100% Honestly, it's probably quicker and easier to do this as well. 

If I drain the overflow box and redo the bulkhead then I have to let it dry for at least 24 hours which means I can’t

run the tank/filters/oxygenation, correct?

Also flow through My 20 gallon refugium will be turned off since the pump will be off.  

I don’t think you have to let it sit for 24 hours. Maybe 30-60 mins to let the glue set up but all your doing is gluing two joints. Don’t think that would cause you an issue.
